Immunohistochemistry Compact Course
Target Group
Technical and scientific staff members with solid knowledge of cell culture and molecular biology
Content/Learning Objective
In the immunohistochemistry the specificity of antibodies is used to show the distribution of specific antigens in the cell or the histological section. Specific antigens only present in some cell types or in particular tissues are ideally suited for this purpose. In this course we will handle:
- processing of tissue (cells, paraffin and frozen section) fixation and deparaffinisation of the tissue
- antigen demasking
- basic of immunochemistry, (diluation of antibodies, incubation, stability, cross-reactivity etc.) and different immunochemically staining methods
- (fluorescent technique and enzymatic staining with chromogens)
- problems as high background, application of controls and false-positive results will be covered in trouble shooting
Main Topics
Preparation of specimens, fixation, immunofluorescence, enzymatic staining, trouble shooting
