Cell Culture Trouble Shooting

Content & Learning Objective

This course provides you with knowledge and techniques to identify and avoid cell culture problems such as slow cell growth, reduced survival after thawing, and contaminations. You will be introduced to documentation and standardization methods as tools to optimize cell culturing. During the practical section, we will analyze the effects of the most common mistakes in routine cell culture, e.g., incorrect protocols and optimal freezing, thawing, and subcultivating cells, as well as mistakes resulting from the mishandling of cell culture media. Furthermore, you will be trained to early recognize contaminations and to avoid them by adopting certain standard procedures.

The theoretical part includes:

  • Basics of sterile working techniques
  • Routine methods in cell culture
  • Components of media and their use
  • Cell counting and viability testing
  • Contaminations in cell cultures
  • Development of a trouble shooting guide
  • Documentation
  • Cell banking

The practical part contains

  • Thawing of cells with and without errors
  • Subcultivation of cells with and without errors
  • Freezing of cells with and without errors

Technical and scientific staff members with cell culture experience.

Teacher

Associate professor Dr. Rüdiger Arnold  teaches at the University of Heidelberg as a lecturer in several life science education programs. Dr. Arnold studied biology in Giessen and worked as a postdoctoral fellow at the Max - Plank - Institute of Physiology and Clinical Research in Bad Nauheim. Afterwards he joined the German Cancer Research Center and habilitated at the medical faculty of the Ruprecht-Karls - University of Heidelberg in immunology where he worked in the area of activation, differentiation, and apoptosis in leukocytes and lymphoma. Between 2011 and 2015 he headed the student research laboratory of the Heidelberg Life - Science Lab at the German Cancer Research Center. He also has many years of experience as a lecturer in training for professional qualifications tumor biology DIW-MTA .
